Below is a picture of my rear window, its like this all the time.

Ive located a leak coming from halfway up the C pillar on the passenger side which is collecting in the boot (almost a pond some days!)

And the de-mister button takes about 30mins to start working
What is the likely solution to this? removing the rear window then re-sealing it??

If so, how much am I looking it?

Isnt that what your insurance if for ? can you not get the glass replaced for a nominal fee?

Wouldnt I lose the NCB?

Nah , i don't think glass is counted towards NCB.

Remember to make sure they fit an MB window :)

Do you mean 30 mins to clear the window or 30 mins before the demister light comes on and stays on? May sound daft but mine used to flash for upto 10 mins before it would stay on, I think it was due to a low battery. New battery and no problems now.
You can get some sealer called seek'n'seal or similar for fixing window leaks, depends how big the hole is or you could try some black silicone sealant if you dont want to claim on the insurance.

I get that happening to mine as well I just put it down to the bad weather and trampling all the damp into the carpets
