having read the handbook I am trying to operate the roof as follows on my 2009 R171 but to no avail.

1 Auto opening on the key. I have tried to remote open the roof using the key. I depress the open button and keep it pressed but the roof still doesn't open?

2 When opened from the inside I am trying to close the rear 1/4 lights. The handbook says press the roof button twice in rapid sucession, but they still don't close

Does the car require dealer programing or am I doing something wronG?

Can't help with remote roof operation (that's one option Angie's SLK doesn't have  ) but AFAIK you cannot raise the rear 1/4 light windows with the roof down, only lower them with the roof up thus giving the car a "windowless coupe" look.

BTW, if you're looking to control wind buffeting with the roof down, then the only solution is the wind deflector. It may cost lots of money for the clear one, but it's very effective.

Edit: I believe that you have to point the remote at the sensor on the driver's door handle to operate the roof as it works only through the infra-red function, not the radio one.

Isn't it a one touch function to operate the roof in the R171 and not continued operation of the switch?

You may just need to check programming with the dealer...

I fitted a vario roof module to my SLK32 which allows one touch roof operation from inside the car to allow me open or close the roof even while the car is moving, and up to speeds of 28mph! (Now thats what I call a wind brake! :D) No more constant operation of the switch, far more convenient.

From outside the car, (in my case), its a succession of 5 quick presses to open the roof using the unlock button on the key fob - to close it, three presses on the lock button on the key fob.

When the roof is up three presses of the unlock button of the key fob lowers the windows and three presses of the lock button raises the windows.

I think you're referring to the SmartTop module, which is an aftermarket accessory.

Vario-roof is MB's name for the standard SLK roof, and in standard operation the switch needs to be held throughout the open/close cycle. This can be done when the car is stationary or at low speed (up to about 5mph or 5km/h - not sure which). Remote operation is an option (from R171 onwards) and if fitted, requires the key to be held close to - and pointed at - the driver's door handle throughout the open/close cycle.
