I have just got a 2012 SLK 250Cdi which I want to connect my iPod to. The audio menu offers USB or Bluetooth Audio but not iPod (like my E Class). The glove box has a USB Port and a 3.5 mm jack but there is no audio slot as in the E Class.

When I plug the iPod into the USB port, it says "Unable to read USB"

Help? :fail

This has been covered many times on the forum.
The iPod cannot be connected using the lead supplied to the usb post. There is a dedicated lead for head unit to iPod IF your car is fitted with either an iPod interface or Media interface. If you have neither (as I suspect), the options are auxiliary lead, male to male (headphone socket to 3.5mm jack) or wireless transmitter to tune the radio in or if there is such a thing, bluetooth dongle for the bottom of your iPod or something like a Denison Gateway.
Or you have the iPod or Media interface fitted………..comand.co.uk

If you have comand or Audio-20 with integrated six disk changer then you could add the media interface.
